Mutations in HLA genes seem to play a crucial role in the pathogenesis of some cases (but not all), likewise silica, aromatic and chlorinated solvents, ketones, trichloroethylene, welding fumes and white spirits exposure seems to contribute to the condition in a small proportion of affected persons. Vitamin D is implicated in the pathophysiology of the disease.

Jun 04, 2013 I visited my rheumy today and he has prescribed plaquenil to help treat my scleroderma. I have limited CREST scleroderma. My symptoms seem to have worsened over the past couple of months, mainly, extremely fatigued & a lot of joint pain. Also, I am noticing that I become much more short of breath than I ever have before. Scleroderma is an autoimmune disease affecting the connective tissue in multiple organs, including the skin, heart, kidneys, and gastrointestinal tract, due to excessive production of collagen, a connective tissue protein. Collagen, a key component important of scar tissue, can in excess lead to hardened and thickened tissue in organs and joints. Plaquenil is the brand name for the prescription drug hydroxychloroquine. It's used to treat and prevent malaria infection, and to reduce symptoms and progression of autoimmune diseases such as.

Typical scleroderma is classically defined as symmetrical skin thickening, with about 70% of cases also presenting with Raynaud's phenomenon, nail-fold capillary changes and antinuclear antibodies. An inverse correlation between plasma levels of vitamin D and scleroderma severity has been noted and vitamin D is known to play a crucial role in regulating (usually suppressing) the actions of the immune system.

  4. I may be wrong I often am! but I think there are more listed side effects for aspirin than there are for plaquenil; and by a very long shot, it would seem to be safer than steroids, especially for scleroderma patients. Basically, plaquenil's effect is to modulate the immune system, which is something that could be quite beneficial for some of us.

A second type of scleroderma is called diffuse scleroderma. This means that more areas of the skin are involved and thickened, but there is a high degree of variability among patients. Skin of the arms, legs, and trunk are more likely to be involved. Anti-malarial drugs, such as Plaquenil hydroxychloroquine, is often used to manage lupus and can also be effective in scleroderma. Plaquenil is thought to interfere with communication between immune cells, which can suppress the damaging effect of an overly active immune response without raising a patient’s risk of susceptibility to infections. Hi San It's great to hear from you again! Sorry to hear that you're worried about being prescribed Hydroxychloroquine Plaquenil. I'm not on this drug myself so can't give you any first hand experience but I know many of our members are using it and a lot of them seem to have had very satisfactory results from it.
